In 1750s South America, Jesuit priest Father Gabriel builds a mission among the Guaraní people above a great waterfall, winning their trust and converting many to Christianity. Rodrigo Mendoza, a slave trader and mercenary who once raided the Guaraní, seeks penance for killing his brother and joins the Jesuits at the mission. When a treaty cedes the mission's land from Spain to Portugal, which permits slavery, the priests must decide whether to abandon the Guaraní or defend them by force, leading to a violent, tragic confrontation with colonial troops.
